Abstract

Transformation of chickpea (Cicer arietinum L.) was achieved using Agrobacterium tumefaciens strain EHA105 harboring binary vector p35SGUSINT containing uidA (GUS) and npt II genes, and pJB90GI containing uid A (GUS) and hpt genes. A strong GUS expression was detected in the putative transformed shoots by the histochemical assay. Transformation was confirmed by the presence of 0.69kb and 0.82kb amplified fragments for npt II and hpt genes, respectively, by PCR analysis. Multiple shoot formation was achieved in cotyledonary notch region on MS medium supplemented with 1.0 mM thidiazuron.
